Abstract

Production, Optimization and Characterization of ?Ž?±-Galactosidase by Bacillus sp. SPE10 and Bacillus sp. SPE15 Isolated from Soy Effluent

This study evaluated α-galactosidase production by two Bacillus strains SPE10 and SPE15 isolated from effluent sample. Various physico-chemical parameters such as pH, temperature, incubation period, and effect of different carbon and nitrogen sources were studied in order to determine the optimum conditions for α-galactosidase production by both the strains. The incubation time of 72 h and 50°C with the presence of raffinose and peptone favoured α-galactosidase production in both the strains. pH 8 and 7 were found to be optimum for SPE10 and SPE15 respectively. Purified enzymes showed molecular weights of 81 and 78 kDa revealed by SDS-PAGE.
