Development and Optimization of RP-HPLC Method for analysis of 5-FU in Human and Rabbit Plasma Samples: Identification and Quantification

In vitro and in vivo experiments revealed that drug loaded cross linked chitosan nanoparticles showed; a sustained release profile of 5-fluorouracil compared to the oral solution of 5-FU due to which Area Under Curve (AUC) was increased two times more folds, as promising site specific targeted site of 5-FU to the colon. A simple, accurate, precise, cost effective and sensitive Reversed Phase HPLC (RP-HPLC) method has been developed and validated then, for determination of 5-FU in human and rabbit plasmas. The current RP-HPLC chromatography system is an isocratic of Agilent technologies series 1200 consisted of a pump and variables.
